Assassin's Creed 3 may hit New York City according to clues we've collected so far. We know Desmond's fight against the templars somehow leads him to America's past. How Desmond is related to this new assassin remains a mystery. Some sleuthing through history, both Desmond's and America's, may answer more of our questions about Assassin's Creed 3.
The final mission in Assassin's Creed: Brotherhood DLC "The Da Vinci Disappearance" has Da Vinci and Ezio in a room similar to the one at the end of the game. In it, lies a mystical statue with a message. Aligning the symbols correctly gives you a list coordinates that read 43°39'18"N, 75°27'42"W. Da Vinci thinks the finding is meaningless, but is he wrong? Spoiler: He totally is. These coordinates lead us to New York.
Desmond's father, William Miles, was viewing this memory from the Animus. He left to find those coordinates, believing them to be another temple.
In Assassin's Creed: Revelations, Juno tells Desmond he must travel to the Grand Temple which was left behind by the First Civilization. The Grand Temple is located in North America, more specifically New York. This temple is where Minerva, Juno, and Jupiter hoped to find a way to save humanity from a solar flare. Their plans failed and the result was the First Disaster, a devastating event that nearly wiped out humanity. Juno, Minerva, and Jupiter foresee a second disaster of this magnitude to occur in 2012.
When Desmond wakes from his coma at the end of AC: Revelations, he finds himself in a van with Shaun, Rebecca, and his dad. The van doors open to reveal the field where the Grand Temple lies; Meaning Revelations ends in New York.
Now that we can all agree Assassin's Creed 3 takes place in New York, let's try to figure out where in the state's history this game is taking place.
An Ubisoft survey from last year asked its participants to choose which setting they found the most compelling.
"The confrontation between British colonists and native Americans during the American revolution of the XVIII century" was one of the options. If we go off of this survey, then we should assume the focus of the game won't simply be about the Revolutionary War, but also the violent confrontations between the British and the Native Americans.
The British base during the 18th century was located in New York City. The British took Manhattan after the defeat of Fort Washington on November 16, 1776. They held the city until 1783. The Great Temple's remains were under British authority, whether they knew it or not.
The Greater New York area belonged to the British during the American Revolutionary War. In turn, the area also became the center of the Patriot center intelligence network. Its leader was none other than George Washington. Nathan Hale, Benjamin Franklin, and John Jay were all involved in this secret origination that specialized in double agents, sabotage, codes and ciphers. Paul "The British are Coming" Revere was one of the first famous “intelligence” operatives.
We don't know a ton about the British occupied New York. That's why this location is ideal for Ubisoft to tell a historical story without running the risk of being inaccurate. The wide central intelligence network created in that area would certainly need an assassin or two.

We're still in the dark about Native American aspect of our new assassin. It's safe he's related to our hero, Desmond. But how?
The Iroquois, also known as the Haudenosaunee, lived in the Greater New York area during the American Revolution. To be fair, they lived there long before those events as well. Iroquois tribes tried to stay neutral once the Revolutionary War started. Eventually they had to take sides. The Tuscarora and the Oneida sided with the colonists but the Mohawk, Seneca, Onondaga and Cayuga stayed loyal to Britain. The Iroquois had previously taken the British side in the French and Indian War several years before. The American Revolution marked the first political split between the six tribes.
Mohawk war chiefs and British allies attacked numerous settlements, destroying villages. George Washington ordered an attack, "not merely overrun, but destroy," the British-Indian alliance. I believe the assassin on Assassin's Creed 3's box art is an Iroquois, most likely an Onondaga and Cayuga, based on the fact that the assassin on the cover is ready to take a tomahawk to the head a of Red Coat. If the survey mentioned before is to be considered, I think these alliance will be at the center of AC3's plot.
I've done my share of internet sleuthing today and I think we're on the right track. What do you think is in store for Desmond?

I'm totally excited for this game, for me being a Native American I'm glad to see a respected developer making a game involving the original people of the US and Canadians are friendly to the NA's up there not many people do that, can't wait for this game to come out so stoked.

Can't wait to see the historical figures from the Revolutionary War time as only Assassin's Creed can portray them and see which ones were Assassins or Templars. Wonder if we'll see George Washington use his Apple of Eden (as seen from The Truth puzzel). Also, wonder if they'll put multiplayer in this one and what new things they'll do with it.

There are so many events that occur in the New England area like the Boston Massacre, Boston Tea Party ( Remind you they dressed up like Native Americans ), Paul Raveer's Famous Ride, and First shot of the Revolution was fired in the Massachusetts ( Lexingtion/Concord ), and the Battle of Saratoga in New York!

For the people who dont think this could happen THINK PLEASE. If you know your history then you know that in the revolutionary war the french helped the patriots (americans) SO maby somehow the desendent of ezio or who ever the person is was in france and a group of assasins came to America to help fight the war against the lobsterbacks!!!! Its amazing what happens when you know your own history haha :)

I haven't seen anyone bring up the Freemasons. Many of the founding fathers were masons and I've seen a few documentaries where people believe the organization was linked to the original Templars. I've always figured they would tie in the Freemasons with the AC lore, so makes sense to take place in the American Revolution. From the documentaries I've seen, it appears that the Freemasons were a group of people who met in secret so they freely discus ideas and issues they have with those in power without fear of being prosecuted by religious or royalty leaders. Sounds more like Assassins then Templars to me.
I'm guessing it will turn out that the masons are a branch of the Assassins in the game and the hero is probably the offspring of a frontiersman or trapper and a Native American (probably from a Canadian tribe, since it is where Ubi is). So he gets trained to fight by his tribe, but is fluent in English language and customs so he can mingle with the crowd just fine in a place like New York. He befriends a Freemason and they start giving him targets to kill. It would be neat if in a future spin-off game they sneak him into London to spy and kill or send him to France to help them with their revolution.
The Illuminati also appear to have the same ideals, so I figured that they would eventually do a game about one or the other secret society, but since they appear to be going forward in time the Freemasons make more sense.

I believe the Assassin is a Colonist, and that the Iroquois are actually a faction similar to the Thieves Guild.

It's possible the Assassin is part native american. After all, Desmond is originally from the Black Hills which were occupied by Native American for quite some time before being taken by the US. Also, it may have some connection as Desmond did travel to New York after fleeing the Black Hills which is where he was captured by abstergo.

I think the Assassin is a colonist and that the Iroquois are an allied faction, similar to the Mercenaries, Thieves, and Courtesans.
